High Resolution Multiplex Optical Imaging of Metabolic Dynamics in Animals

Understanding the dynamics of metabolism in a multicellular organism is essential to unraveling the mechanistic basis of many biological processes. Stimulated Raman Scattering (SRS) can generate chemical specific imaging with high resolution, deep penetration of depth, multiplex, chemical selectivity, 3D volumetric and quantitative capability.

In this webinar hosted by the Microscopy and Optical Coherence Tomography Technical Group, Lingyan Shi from the University of California, San Diego, will present on the development of a new method that combines deuterium oxide probing and SRS to visualize metabolic dynamics in living organisms. Within the broad vibrational spectra of newly synthesized carbond deuterium (C-D) bonds, they discovered more than 20 different lipids subtypes-, protein-, and DNA-specific Raman profiles and develop hyperspectral detection methods to obtain various macromolecular multiplex imaging.

This technology platform is non-invasive, universal applicable, and it can be adapted into a broad range of biological studies such as neurodegeneration, aging, homeostasis, tumor progression, etc. Dr. Shi’s lab has applied this method to study the diet regulated metabolic dynamics in animals during aging processes, the quantitative lipid and protein turnover rate, and the intra-cellular metabolic heterogeneity.

About the Presenter: Lingyan Shi, University of California, San Diego

Dr. Lingyan Shi is Assistant Professor in the Department of Bioengineering at UCSD. Her lab focuses on developing and applying ultrafast laser scanning optical imaging and spectroscopy imaging platform to investigate cellular metabolic dynamics in living organisms. She developed a breakthrough platform that combines deuterium oxide probing and SRS (DO-SRS) microscopy to visualize metabolic activities in situ, and discovered a Golden Window for deep brain imaging which suggests that one should put both excitation and the emission wavelength in the optical tissue window. Dr. Shi has been awarded 6 patents as a co-inventor. She is an OSA Senior Member. She won Blavatnik Regional Award for Young Scientist in Physical Sciences & Engineering in 2018, and Hellman Fellowship Award in 2021. She received “Rising Star Award” by Lasers Focus World, “Faculty Fellow 2021” by Faculty Fellow program Israel, and “Advancing Bioimaging Scialog Fellow” by RCSA and the Chan Zuckerberg Initiative in 2021.
